blisters will happen on the trail. moleskin and bandaids will help but you're much better off trying to prevent blisters. if they occur, you'll likely be walking through whatever pain and raw skin you have. its a bit painful and gross but i've seen people go hundreds of miles like this. when it comes down to it, blisters are more likely to be an annoyance than a problem.
the best way to prevent the problem is to wear the shoes before the trip. better yet, wear the shoes with a loaded pack. go for a couple day hikes. a heavy pack changes the way your body carries weight which can change the pressure points inside your shoes.
i'm unfamiliar with the trail but if there's a lot of elevation change then you might find a tall building where you can climb stairs. up AND down.
